Minamino Yoko got her start in show business after she won an audition when she was a second year student in high school. She first become an actress in 1984, debuting the television dorama Meimon Shiritsu Joshi Koukou. Then a year later, she debuted as a idol singer with the single "Hazukashi Sugite".

Minamino wasn't getting a lot of promotion and popularity until she landed the lead role as Saki Asamiya for the dorama Sukeban Deka II. This role made her a overnight sensation and further her popularity among Japan. This was seen in her idol singer too as her 1987 single, "Rakuen no Door", topped the charts and sold many copies. She would go on to score nine #1 hits during her singing career. At the end of the decade, she was named one of the Four Queens of J-Pop, along with Kudo Shizuka, Nakayama Miho, and Asaka Yui.

In 1988, she portrayed a double role in the dorama Takeda Shingen. At that same time, her Bromide sales set a record, reaching #1 for all Bromide sales at that time. Her image was also used on postal stamps in Grenada. But, all this popularity was making her a bit stressed when one of her memorable moments of her career was when she forgot her lyrics to the song "Aki Kara mo, Soba ni Ite" when performing on the music program Music Ten. She broke down crying and asked for a restart, which the program let her do.

In 1989, Minamino was featured as a charity report for the special program 24 Hour TV, reporting about the troubles of Cambodia, and singing the theme song for the special Omoi no Mama ni. She also changed agencies in the same year but didn't make the public very happy about it. The media bashed Minamono for the move and her idol popularity started to take a downturn.

Starting with the 90s, Minamino focused more on her acting career then her idol singing career. In 1992, she made her first nude appearance in the movies Kantsubaki and Watashi wo Idaite, Soshite Kiss Shite. Her roles in these two films earned her the Best Lead Actress at that year's Nihon Academy Taishou. Three years later, to celebrate her ten years in the spotlight in 1995, Minamino starred in various stage appearances like Yuzuri Ha no Ido. She also changed agencies to Sweet Basil before the decade was up in 1999.

In 2002, she starred in the noon dorama Ikkakusen Kin Yume Kazoku, which garnered high ratings for a single noon dorama. Three years later, to celebrate her twenty year anniversary since first debuted as a single, Minamino released a DVD box set, a new compilation box set, and a new single, a self cover of 1987 single "Haikarasan ga Oru". She also announced on the music program Utaban that she would restart her music career after a 14 year hiatus. Before the year was up, Minamino also debuted as a radio personality.

In 2008, she became the public relations ambassador for her hometown of Itami.
